**Background**
The serotonin 5-HT7 receptor is a G protein-coupled receptor widely distributed in the central nervous system and peripheral tissues, including the immune system. This receptor plays a critical role in regulating cognitive functions, such as memory consolidation and learning, as well as modulating inflammatory responses and pain sensitivity. Due to its involvement in reversing amnesia and enhancing T-cell activation, the 5-HT7 receptor has become a significant target for developing therapeutic agents for cognitive impairment and immune disorders. In this context, we will introduce a potent and selective 5-HT7 receptor agonist – AS19.
**Definition**
AS19 is a potent and selective 5-HT7 receptor agonist with an IC50 value of 0.83 nM and a Ki value of 0.6 nM. According to the AS19 description, this compound exhibits high selectivity for the 5-HT7 receptor over other serotonin receptors, including 5-HT1A, 5-HT1B, 5-HT1D, and 5-HT5A.
**In Vitro and In Vivo Studies**
The AS19 biological activity has been demonstrated across various experimental models. In terms of AS19 In Vitro studies, the compound has shown significant effects on the immune system; specifically, the proliferation of T-cells from parachlorophenylalanine-treated mice was significantly reduced, but the addition of AS19 (1 μM) completely restored T-cell proliferation after 48 hours. Furthermore, AS19 has been shown to inhibit mechanical hypersensitivity secondary to capsaicin sensitization in mice.
Regarding AS19 In Vivo applications, the compound has demonstrated a strong capacity to enhance cognitive function. In an autoshaping Pavlovian/instrumental learning task using 12-week-old male Wistar rats, subcutaneous injections of AS19 at dosages of 0.5 mg/kg, 1 mg/kg, 5 mg/kg, and 10 mg/kg for 24 hours significantly improved memory consolidation. Additionally, AS19 has been reported to reverse amnesia induced by Scopolamine or Dizocilpine. For researchers requiring specific AS19 technical information, it is noted that the compound has a molecular weight of 283.41 and a chemical formula of C18H25N3. In conclusion, AS19 is a highly selective 5-HT7 receptor agonist that holds promise for research into memory enhancement and immune modulation.
