Ocessed for microscopy as previously described (Yong-Gonzales et al., 2012). The exposure times used for Tub1-GFP and Hoechst were two and 0.two s respectively. All imaging was captured on an Axio Imager microscope (Carl Zeiss, Jena, Germany) using a 100objective lens (numerical aperture 1.four). From 8 to ten Z-sections with a 0.5-m step size had been taken to cover the entire yeast cell. Unpaired Student’s t test was employed for statistical evaluation.ACKNOWLEDGMENTSWe thank Maria Longhese (Universitdegli Studi di Milano-Bicocca, Milan, Italy) for the TEL1-hy909 allele and David Toczyski (University of California at San Francisco, San Francisco, CA) for the strain containing the LacI-GFP fusions of Ddc1 and Ddc2; Koyi Choi for her aid in generating some of the strains; and members in the Zhao lab for comments around the manuscript.
